Amazon kindle you can create your account on Amazon Kindle. If you already created an Amazon account for any purposes then you can just login to your account you don't need to create another one.
Don't over think about your ebooks are in form of Kindle format or not. Amazon will convert your ebooks according to their needs or their format. You just don't need to worry about Kindle format. You just write your ebook properly.
After you sign up to Amazon Kindle, Amazon will take you to your book storage space, here you can find all your books that are ready for sale. At first obviously the book space will be blank but not for long. It'll start filling up once you start to uploading your books on that platform.
It will take only few more steps for your eBook to be on Amazon Kindle direct publishing for sell. It's really easy to do that actually.
When you open you Kindle direct publishing account you'll see a tab "add new title" at the top left side if your screen.

When you first start to fill up all those section the first title would be 'enter your books details', and there is an option you'll see called ISBN. Full form of ISBN is international standard book number. And giving ISBN number is optional that means it's your choice that you want to give the number or not. But most of the time bloggers or publishers don't have that number.
Most of the article I when I first started selling by eBooks on Amazon Kindle, they said you needed an ISBN number to publish books on Amazon Kindle. I don't think I would be even able to published my ebooks on Amazon Kindle and get my books listed on that site.

You don't necessarily need an ISBN number to publish your eBook on Amazon Kindle or even sell them online. So my advice to you that just leave that column blank.
But I would say if your book is published by your self then give your own on that ISBN column.
Then you'll need an perfect image for ebook to use as cover photo. And it's a good idea to spend some times to create that cover image because that image will represent your eBook on Amazon sale page. If you feel that you don't have the capabilities to make a good cover photo for your ebook then can hire someone to make that cover for you.

You can use free learncher site like fiverr to hire a good graphic designer. There are lot of good quality and high skills people on site Fiverr who can design the perfect cover photo for your ebooks to be display on Amazon sale page. And it's all can done as little as $5.
Amazon preferred a JPEG or TIFF type format images for their site so it important for you to upload that type of image on Amazon sale page as your eBook cover photo. Your cover image height/width ratio should be 1:6 and atleast 1000 pixels.

Amazon give you two royalty option to choose from which are 70% and 30%. Let say if you fixed your eBook price $10 for sale and your option is 70% royalty then you will get $7 once your ebook sold and the rest $3 will amazon keep as commission. If you set your book price below $2.99 then you are eligible for 35% royalty option.
